Human stomach grown in vitro 06.11.2014

Researchers at Cincinnati Children's Hospital Medical Center (USA) have managed to grow a human stomach in a test tube. True, in size it does not exceed 3 mm in diameter, however, despite its diminutiveness, it largely repeats the structure of a "natural" organ.

James M Wells and colleagues used human pluripotent stem cells, from which any type of body cell can be obtained. The stem cells themselves were of two types: natural, obtained 15 years ago from a human embryo, and artificial, or induced, obtained as a result of molecular reprogramming of specialized skin cells. Currently, the use of stem cells from an embryo is associated with many legal problems, so induced pluripotent stem cells are more often used in such studies. However, cells taken from an embryo many years ago, thanks to special storage methods and due to their own immortality (they can potentially live and multiply indefinitely), can also be used in experiments of this kind.

The essence of the task was to guide stem cells through several successive stages of stomach development. First, the so-called internal germ layer, or endoderm, was formed, which is formed at the earliest stages of embryo development. Endoderm cells, although somewhat losing their "omnipotence", can still give rise to a variety of organs, not only the stomach, but also the lungs, liver, and pancreas.

At the next step of specialization, such an artificial endoderm received a signal to turn into a tube - a prototype of the gastric sac. (A signal should be understood as another portion of special proteins that bind to cell receptors and change the activity of genes.) Actually, this was the main result of the experiment: a flat cellular structure was turned into a three-dimensional one. Such a transformation occurs with an ordinary embryo, but in laboratory conditions it is incredibly difficult to reproduce the stage of transition from 2D to 3D.

Such a transition has become one of the main tasks in modern developmental biology, which has been solved in a variety of ways. This is especially true of complex organs, with a complex internal structure, consisting of several types of cells and permeated with blood vessels.
