Stem cells from a test tube 23.11.2012

Canadian and Italian researchers have found a "master control gene" for human blood stem cells. Scientists have found that by turning this gene on and off, an efficient way to grow blood stem cells suitable for clinical use can be obtained.

According to Dr. John Dick, Canadian Professor of Stem Cell Biology, Lead Scientist at the Center for Restorative Medicine and the Ontario Cancer Institute (OCI), and Professor in the Department of Molecular Genetics at the University of Toronto, the scientists' discovery introduces a new paradigm for managing human blood stem cells. The results of the study will soon be published in the journal Cell Stem Cell.

"For the first time in human blood stem cells, we have discovered that a new type of non-coding RNA called miRNA provides a new tactic to control these cells, opening the door to growing them for therapeutic purposes," said Dr. Dick. In 2011, Dr. Dick isolated human blood stem cells in their purest form, as a single stem cell capable of restoring the entire circulatory system.

According to OCI lead author Dr. Eric Lekman, the research team has removed the master control gene - microRNA 126 - that normally controls hundreds of other genes, inhibiting their activity, which in turn leaves the stem cell dormant in an undivided state. The method consisted in creating an excessive number of microRNA-126 binding regions in stem cells using a specially designed virus.

"The virus acts like a sponge, erasing a specific miRNA within the cell. This allows the suppressed genes to become more active, after which we see long-term growth of stem cells, without depletion or malignant changes," said Dr. Lekman. "We have shown that by removing miRNAs, you can grow stem cells while keeping their properties unchanged. This is the key to efficiently growing stem cells for use in patients."
