Focus Description:

The performer shows the audience an empty vase. His assistant brings a tray onto the stage, on which stands a decanter of water. The magician takes a decanter, and in its place puts a vase and fills it with water. Then he covers the vase with a handkerchief and lifts it from the tray with both hands. The assistant takes hold of the edge of the tray with one hand and carries it backstage.

And the performer with a vase in his hands goes to the audience. Having reached the first rows, he throws a vase with a handkerchief into the air, and then catches the handkerchief - the vase was no longer in it. She disappeared. The performer shows the scarf that covered it to the audience from both sides - there is certainly no vase in it.

Focus secret:

Here, all the props - a vase, a scarf and a tray - have secrets. Let's get acquainted first of all with the device of a vase. It doesn't have to be glass. For a trick, it is better to take an aluminum or enameled bowl that has the shape of a deep plate. In order for it to look like a vase, the bowl must be painted accordingly. A through hole is drilled in the middle of the bottom of the bowl. In the left half, on the outer side of the bottom, two nails 1 cm high with caps are soldered or riveted (this is better) (Fig. 21).

The tray is good to take with a handle. The tray field should be double, consisting of two sheets with tightly closed edges. A hole is drilled in the center of the top sheet. To the left of this hole, closer to the handle of the tray, two more round holes are drilled, turning into narrow slots. The vase-bowl is placed on a tray so that the central holes in the tray and the bottom of the vase coincide, and the heads of the nails easily enter the corresponding holes in the bottom of the tray (the narrow part of the holes in the tray should be equal to the diameter of the base of the nails soldered to the bottom of the vase).

The handkerchief should be of a light, colorful fabric and slightly larger than the size of the tray. In the center of the scarf, a wire circle is sewn in with a diameter equal to the diameter of the edges of the vase.

The vase can be placed on the illusion table in advance. When starting a trick demonstration, the performer asks his assistant to bring a carafe of water. When the assistant takes out a decanter on a tray, the performer removes it from the tray, and puts a vase in place of the decanter. As soon as the heads of the nails enter the hinges of the tray, the magician pours the water into the vase. Water from the vase will immediately go through the hole into the tray. After that, the performer covers the vase on the tray with a handkerchief, but instead of the vase, he lifts the handkerchief with wire. And the assistant at this time takes the tray by the handle and turns it towards the audience so that they do not notice the vase. When the performer, handkerchief in hand, approaches the auditorium, the assistant with the tray retires backstage. Approaching the front rows, the performer throws up a handkerchief and catches it, and then shows it to the audience from both sides.

Scientists have done the impossible and completely reworked the E. coli genome, eliminating all unnecessary from it and replacing the original genes with their synthetic counterparts.

With all the great diversity, life on Earth uses the same "language" - DNA. A handful of conventional chemical "letters" are used to create dozens of three-letter sequences, each of which conveys a specific set of information to protein structures. The four letters of non-glutinous acids - adenine, cytosine, guanine and thymine (A, C, G, T) - can be combined into 64 combinations of three-letter "words", the so-called codons.

Modern life forms are represented by only 61 codons that form 20 amino acids. The other three are punctuation marks of sorts, denoting the endpoint of a particular gene's pattern in a continuous chain. Thus, our genes often use several different fragments to represent the same trait. This creates a huge redundancy of information, but there are good reasons for that. In nature, this allows the body to quickly adapt to environmental changes, but is it possible to keep the number of codons to a minimum under controlled laboratory conditions?

To find out, a research team from the University of Cambridge studied the entire genetic code of an E. coli strain and isolated each time one of three different codons appeared. Two of them mean the amino acid "serine", and the third plays the role of a stop codon. Then each of these triplets was replaced by one of four other codons that also code for serine, and even the stop codon was replaced by one of two analogs.

On paper, making all these edits (and there are about 18 of them) looks as simple as auto-replacing one word with another in an electronic document. But in practice, this is extremely painstaking work, since the researchers needed to collect a chemical copy of the edited genome and replace the original with it without killing the living organism. The team did this in stages, and after replacing each segment, the scientists were convinced that the bacteria continued to function as before. Surprisingly, it worked!

As a result, the Syn61 variant turned out to be the most viable - with it, the cells are visually longer and at the same time multiply 1,6 times slower. That being said, the "edited" E. coli appears to be healthy and works with the same range of proteins as the original version. For researchers, this is very good news, convincingly proving that genetic engineering is capable of literally replacing wildlife with synthetic equal nature to the extent that a person needs it, while preserving life itself.
